Part:BBa_K2754001:Experience

The part was tested by measuring absorbance and fluorescence using a Tecan plate reader. The absorbance was measured at 590nm and the fluorescence at the wavelengths: emission=583nm and exitation=556nm. We have measured it in parallel with the already existing biobrick BBa_J04450, to get comparable results.

The first time we measured the biobrick, we used a transparent 96 well plate and added Isopropyl β-D-1-thiogalactopyranoside (IPTG) when the absorbance level reached 0.8. IPTG was added in concentrations of 0 mM, 0.5mM, and 1.0mM.

The second time we tested the RRvT biobrick, we decided to use black 96 well plates. We also added IPTG from the beginning.

The third run, we adjusted the LB-media to a pH=5.0 to see how this affected the fluorescence.
